Just watched this movie today, loved it. Thank goodness that it wasn't as weird as Being John Malkovich or Adaptation, although it did have it's moments of gratuitous weirdness. Jim Carey deserves an award.
Patrick: Mary hates me. I've never been popular with the ladies. Stan: Maybe if you stopped stealing their panties.
Clementine: Drink up, young man. It'll make the whole seduction part less repugnant.
Clementine: You're not a stalker, or anything, right? Joel: I'm not a stalker. YOU'RE the one that talked to me, remember? Clementine: That is the oldest trick in the stalker book. Joel: Really? There's a stalker book? Great, I gotta read that one.
Obviously, I would immediately choose a few past events and people to erase from the old memory bank. The logical argument would be that doing so would affect me in a negative manner because, after all, who we are is based on what we've experienced. However, the whole 'what doesn't kill you makes you stronger' rationale sounds like a victim's rationale to deal with guilt or regret.

There are plenty of painful memories I'd keep (lessons learned, emotional growth, etc.), though there are a handful I'd erase for sure.
